In this scientific poster, first presented at ISSCR 2024, CRISPR-Ready ioMicroglia are characterised, including cell morphology and gene expression profiles, functional Cas9 expression, single gene knockout efficiency, and applicability to pooled CRISPR screens.
Microglia play a pivotal role in maintaining neural health, defending against pathogens, and maintaining the brain’s functional equilibrium. Leveraging bit.bio’s deterministic programming technology, we introduce CRISPR-Ready ioMicroglia for CRISPR/Cas9-based knockout screening.
In this poster, you will explore data showing:
- How CRISPR-Ready ioMicroglia have been engineered to constitutively express Cas9, which is functional from day 1 post thaw, and retain all defining features of ioMicroglia, such as key markers expression and cytokine secretion profile.
- How the cells demonstrate >84% knockout efficiency at protein level using a guide RNA (sgRNA) targeting beta-2 microglobulin (B2M).
- The cells applicability to knockout screens, including data from a screen to uncover genes involved in microglia activation in response to stimuli.
