MEA co-culture of hiPSC-derived ioGlutamatergtic Neurons with rat cortical astrocytes

Introduction

This protocol describes the plate preparation and co-culture of wild-type ioGlutamatergic Neurons and rat astrocytes for analysis using the MaxTwo High-Density Microelectrode Array (HD-MEA) System (MaxWell Biosystems AG). Optimisation was performed on MaxTwo 6-Well Plate and 24-well plate formats. The protocol is also applicable to any of the ioGlutamatergic Neurons disease model cells.

When co-cultured with astrocytes, glutamatergic neurons show enhanced maturity and synaptic activity. Astrocytes support stable network activity and minimise neuronal stress and cell death, enabling MEA assays over extended periods.
