CRISPR interference (CRISPRi)-Ready ioGlutamatergic Neurons constitutively express catalytically inactive Cas9 nuclease (dCas9) fused to a transcriptional repressor for gene repressions, pooled or arrayed CRISPR interference screens.
The cells mature rapidly into functional glutamatergic neurons after revival in the recommended, fully disclosed medium. They are delivered in a convenient cryopreserved format and provide a homogeneous and reproducible model for human excitatory neurons.
Guide RNA delivery
Guide RNA can be delivered from day 1 to day 11 post-revival, and experimental readout is recommended from day 5 post-guide delivery. The user manual includes protocols for guide RNA delivery and flow cytometry readout.
- Phase 1 - Stabilisation (day 0 to 4)
The cells are revived at the user’s laboratory using the recommended medium supplemented with doxycycline and DAPT for sustained induction. - Phase 2 - Maintenance (day 4 to 11)
The maintenance of the CRISPRi-Ready ioGlutamatergic Neurons can be extended beyond day 11, according to the assay requirements.
