CRISPR-Ready ioMicroglia
cat no ioEA1094 Early Access | Male human iPSC donor-derived microglia expressing Cas9 for rapid gene knockout generation

CRISPR-Ready ioMicroglia rapidly mature into functional microglia after revival in the recommended medium. These cells constitutively express Cas9 nuclease so users can perform single gene knockouts, pooled CRISPR screens or arrayed CRISPR screens. CRISPR-Ready ioMicroglia are delivered in a convenient cryopreserved format and provide a homogeneous and reproducible model of human microglia.

The user manual provides full details on the generation of these cells using a simple three-phase process and includes protocols for gRNA delivery by either lentivirus transduction or lipid-based transfection.

  • Induction
Carried out at bit.bio before distribution.
  • Phase 1 - Stabilisation (24 hours)
CRISPR-Ready ioMicroglia are revived at the user’s laboratory using the recommended medium.
  • Phase 2 - Maturation (9 days)
CRISPR-Ready ioMicroglia are matured by the user in the recommended medium. Cells are ready to use from day 10.
  • Phase 3 - Maintenance
Depending on assay requirements, CRISPR-Ready ioMicroglia can be kept over different lengths of time in the maintenance medium.

The guide RNA can be delivered with lentiviral transduction from day 1 to day 18 post-thaw or by lipid-based transfection at day 10 post thaw.
