Protocol overview
Introduction
This protocol describes the plate preparation and co-culture of ioMotor Neurons and rat astrocytes for analysis using the MaxTwo High-Density Microelectrode Array (HD-MEA) System (MaxWell Biosystems AG). Optimisation was performed on MaxTwo 6-Well Plate. The protocol is also applicable to any of the ioMotor Neurons ioDisease Model Cells.
When co-cultured with astrocytes, hiPSC-derived motor neurons show enhanced maturity and synaptic activity. Astrocytes support stable network activity and minimise neuronal stress and cell death, enabling MEA assays over extended periods.
